We seek to understand how cells regulate their genes and how these circuits change to alter cell fate decisions in normal development and disease. To accomplish this goal, we develop new experimental and computational tools that measure gene regulation across single-cells. In this talk, I will present our efforts to build sequencing and imaging technologies to measure chromatin accessibility, gene expression and genome structure. Further, I’ll discuss how we’ve used these tools to find chromatin accessibility mechanisms of lineage priming and epigenetic memory of genome structure.
